One of the serious tools that is used for mastering is the Spectrum Analyzer, which displays the energy at the different frequencies of the audio spectrum. Izotope's Ozone 3 software displays the spectrum graph in the same window as the graph that shows you the frequency response of the equalization you are using, a very useful arrangement. On this grid you can also display "snapshots" of the tune's spectrums at different points in the tune, display spectrum guides, and use these to have the software calculate the EQ settings needed to get the tune's spectrum to match a target.
The bottom line is that these very powerful tools (spectrums, guides, matching EQ), must be used to the degree that they make the resulting master sound good. The resulting spectrum you have is determined by a lot of factors, such as what instruments are playing, the parts they're playing and the EQ you used on the individual tracks in the mix. There is no automated system that can take all of this into account. These are simply tools and guides to you listening.
